In "Toi et moi sommes plus que des amis.", is the "you" supposed to be 'tu' (a subject pronoun) instead?

"Toi et moi" is correct - this is different to usage in English. Proper names (Martin), nouns (ma mère) or stress pronouns are used for the pronouns in talking about people in these constructs.
